More a TV thing than movies, but William Shatner as James T. Kirk. As my Dad always would say "Shatner isn't a good actor, but he's a great Kirk"
 
How could we overlook the epic performance that Bo Derek gave playing the role of whats-her-name in "10"?

Jack (RP McMurphy), Louise Fletcher (Nurse Ratched), Will Sampson (Chief Broom), Danny DeVito, Brad Dourif, Christopher Lloyd.....hell, all of the "mental defectives" in "One Flew Over the Cuckoo's Nest". Perfect cast in a great film.

First, Robin Williams was a perfect Garp.

Squib, that is really weird. My son is going to attend Alabama. His mother's maiden name was Squibb. When she got a little concerned about the tornado that killed close to 300 in Tuscaloosa, I reminded her of the scene where Garp bought the house because it was "predisastered".

Rickman was perffectly cast as the voice of Mufasa's brother in The Lion King.

Wayne was the perfect Rooster Cogburn.

Eastwood was the perfect Josey Wales.

Hoffman in The Graduate deserves mention

BUT, the best pairing ever was Paul Newman and Joanne Woodward in The Long Hot Summer along with Lee Remick, Anthony Franciosa, Angela Lansbury and Orson Welles.

While I thought Pirates 4 was mailed in (but still worth a look), Johnny Depp was excellent as Jack Sparrow.

Belushi as Bluto?

I do understand the distinction between best performance and best casting.
 
Val Kilmer as Doc Holliday. I doubt the authenticity of the portrayal but nevertheless, Kilmer's adaptation made that film what it was imho.
