Which casino is the nicest? Best poker room?

IMO,

Nicest: Sam's Town. I get comped pretty easily.

Best Poker Room: Binion's by far.

We used to always stay at Horseshoe, until they stopped sending us comped rooms.

Last time we stayed at Sam's Town, and while it was okay (and we get room comps in the mail since then), we liked the casino next door at El Dorado much better. The quality of everything there seemed much more Vegas like than anything else I've seen in Shreveport. i.e. bigger poker room, more table games, more slots, more video poker, HD TV's everywhere, quicker buffet comps, hotter waittesses, etc...
