Reformatting master drive - remove slave?

I want to start fresh with this box but I'd rather not open it up again. Do I need to take out the slave drive in order to make damn sure I don't screw anything up on it during a reformat/reinstallation of windows? I imagine it can be done without removing it, but I really really don't want to screw this up.

I plan on backing up all my important stuff and moving all my songs/videos to the slave drive and then just wiping my C drive and starting over. Any other things I need to be concerned about?
